Στη μυθολογία, η Δονούσα έπαιξε σημαντικό ρόλο στην εξέλιξη της σχέσης του Διονύσου με την Αριάδνη, αφού ο θεός την επέλεξε για να μεταφέρει εκεί την εκλεκτή του και να την προστατέψει από τον Θησέα, που εν τω μεταξύ, την είχε εγκαταλείψει στη Νάξο. Σίγουρα πάντως κατά τους Ρωμαϊκούς χρόνους το νησί υπήρξε τόπος εξορίας ενώ τον Μεσαίωνα χρησίμευε ως πειρατικό ορμητήριο.
Η Δονούσα κατοικήθηκε ξανά το 18ο αιώνα από κατοίκους της Αμοργού. Στη Μεσσαριά βρίσκεται ο αρχαίος οικισμός της Δονούσας στην περιοχή Βαθύ Λιμενάρι ο οποίος χρονολογείται από τον 8ο-9ο αιώνα π.Χ.

Η πρόσβαση στο νησί γίνεται με την Blue star Ferries απο το λιμάνι του Πειραιά ( www.bluestarferries.gr ) καθολη την διαρκεια του έτους.
